Muna’s oral Alzheimer’s candidate shifts age-associated markers in Phase 1

Why it matters: Alzheimer’s has no cure, and the EU-approved antibodies lecanemab and donanemab only slow decline in selected early-stage patients with confirmed amyloid and no more than one copy of the ApoE4 gene variant, while requiring infusions and monitoring for brain swelling and bleeding. Rather than targeting amyloid deposits, Muna’s pill aims to strengthen microglia, the brain’s immune cells, through TREM2, a receptor whose impaired function raises Alzheimer’s risk.

Why it matters: With this round, Arctic is backing AT-002 and AT-003, treatments that could address gaps in the care of patients with autoimmune diseases such as uveitis, an eye inflammation that can threaten sight and whose steroid treatment increases the risk of glaucoma or cataracts. Meanwhile, its clinical-stage drugs target acne (AT-004) and dementia (AT-001), starting with HCCAA, a rare, fatal inherited disease that causes brain hemorrhages and dementia.

Evaxion reports durable responses; Novartis halts kidney trial; AstraZeneca signs AI deal

Evaxion reports sustained cancer vaccine responses at three years: Denmark’s Evaxion said all six patients in a Phase 2 extension maintained their responses without tumor progression after three years. The advanced melanoma patients received personalized peptide vaccine EVX-01 alone during the third year, following two years alongside Keytruda. The original 16-patient trial reported a 75% response rate with 25% of patients obtaining a complete response. The durability signal is encouraging, but the extension included only six of the 12 responders. Further findings will be presented at ESMO on October 24. Evaxion’s shares jumped 21.6% on Monday following the company’s announcement.
