世界农化网中文网报道:近日,Plant Biotechnology Journal杂志在线发表了由先正达北京创新中心梁大伟团队撰写的"A robust framework for maize elite line genome editing through enhanced HI-Edit via LbCas12a activity optimization"论文。该研究工作通过系统优化,成功实现了LbCas12a介导的玉米HI-Edit(单倍体诱导耦合基因编辑);并通过优化Cas12a蛋白在生殖细胞中的表达水平、蛋白稳定性及单倍体诱导过程中环境温度,将该技术应用的核心瓶颈—玉米单倍体编辑效率(HER)从平均不足2%提升至最高33%。研究团队报道的这一套基于保守生物学过程HI-Edit技术策略,为玉米优良种质的直接基因编辑改良提供了明确的技术方案和路径,有望彻底改变传统育种中优良品种难转化、改良耗时长成本高的局面。
